(英) Exploration of proteins contributing to the amitrole-resistance in Escherichia coli
(英) Amitrole is a kind of environmental hormones and inhibits the bacterial growth. It was reported that the chemical mutagen treated Escherichia coli (E.coli) cells exhibit the resistance against amitrole. However, much information on the mechanism and proteins contributing to this resistance were not shown. Therefore, in order to elucidate the mechanism on the amitrole resistance in E.coli, we prepared the E.coli JM109 mutant strain by treating with MNNG, and then selected the amitrole-resistant E. coli strain. The obtained mutant strain was possible for proliferating in the M9 minimal medium containing 2 mg/ml amitrole (AT2 medium). Therefore, we examined the expressed proteins in mutant strain cells cultured under the various conditions. As result, it was suggested the expression of 22kDa protein (P1) was suppressed in the M9 minimal medium containing amitrole. Amino acid sequence analysis of P1 revealed that it might be alkyl hydroperoxide reductase C22 protein. On the other hand, we found the expression of the other protein (P2) was increased when cultured in AT2 medium. Furthermore, we explored the amitrole binding proteins in E.coli mutant cells by amitrole affinity chromatography. And then, five proteins (P3-P7) were selected as candidates of amitrole binding proteins. Further investigations about these proteins (P2-P7) are now undertaken.
